Abstract

Transfection of HT-29 and Hep-2 cells with cDNA of the IL-4 receptor (IL-4R) was demonstrated to result in increased number of functional IL-4R being expressed on the surface of the cells. When exposed to IL-4, marked differences in the percentage of relative cell viability were detected in the transfected HT-29 (HT-29t) and Hep-2 (Hep-2t) cells as compared to their untransfected counterparts. While a significant decreased in the percentage of relative cell proliferation was observed in the HT-29t cells, the percentage of relative cell proliferation was significantly increased in the Hep-2t cells. This confirmed the antagonistic effects of IL-4 on different types of cells. In general, our data also demonstrated that the increase of IL-4R expression rendered HT-29 and HEP-2 cells to be more sensitive to IL-4 and that lower concentrations of IL-4 were required to exert its effect on the proliferation of the cells.
